The following takeoff procedures are our standard ways of departing in a 4-ship (i.e division). All the different procedures can also be applied to (and are recommended for) 2-ship flights (i.e sections) simply by ignoring the #3 and #4 instructions.
Only 2 aircraft should line up on the runway together. #3 and #4 should wait just short of the runway (with #3 past the usual hold point). As soon as #2 has started rolling, #3 and #4 may line up or perform a rolling takeoff. The reason for this is avoid exposing aircraft for jet blast (not simulated in DCS, but we try to respect it).
As a flight lead, you may deviate from these ways and perform any kind of non-standard takeoff, if there's a good reason for it (for instance mixed flights, uneven/unusual loadouts, extreme winds, special exercises). However, in order to establish common standards, the recommendation is to use these whenever possible.

Example
